Therapeutic Targeting of RNA Splicing in Cancer

نویسندگان

چکیده

RNA splicing is a key regulatory step in the proper control of gene expression. It highly dynamic process orchestrated by spliceosome, macro-molecular machinery that consists protein and components. The dysregulation has been observed many human pathologies ranging from neurodegenerative diseases to cancer. recent identification recurrent mutations core components spliceosome hematologic malignancies advanced our knowledge how alterations contribute disease pathogenesis. This review article will discuss current understanding aberrant regulation drives tumor initiation progression. We also therapeutic modalities highlight emerging technologies designed target for cancer treatment.

Role of Aberrant Alternative Splicing in Cancer

Alternative splicing can alter genome sequence and as a consequence, many genes change to oncogenes. This event can also affect protein function and diversity. The growing number of study elucidate the pathological influence of impaired alternative splicing events on numerous disease including cancer. Here, we would like to highlight the significant role of alternative splicing in cancer biolog...

Therapeutic targeting of c-KIT in cancer.

INTRODUCTION Mutated forms of the receptor tyrosine kinase c-KIT are "drivers" in several cancers and are attractive targets for therapy. While benefits have been obtained from use of inhibitors of KIT kinase activity such as imatinib, especially in gastrointestinal stromal tumours (GIST), primary resistance occurs with certain oncogenic mutations. Furthermore, resistance frequently develops du...

Alternative RNA splicing: contribution to pain and potential therapeutic strategy

Since the sequencing of metazoan genomes began, it has become clear that the number of expressed proteins far exceeds the number of genes. It is now estimated that more than 98% of human genes give rise to multiple proteins through alternative pre-mRNA splicing. In this review, we highlight the known alternative splice variants of many channels, receptors, and growth factors that are important ...

Therapeutic Targeting of Cancer Stem Cells

Recent breakthroughs in translational oncology are opening new perspectives for the treatment of cancer. The advent of targeted therapies has provided the proof-of-concept to selectively turn-off deregulated oncogenic proteins, while the identification and validation of predictive biomarkers of response has allowed to improve, at least in some cases, their performance. Moreover, a subpopulation...

Design, simplified cloning, and in-silico analysis of multisite small interfering RNA-targeting cassettes

Multiple gene silencing is being required to target and tangle metabolic pathways in eukaryotes and researchers have to develop a subtle method for construction of RNA interference (RNAi) cassettes. Although, several vectors have been developed due to different screening and cloning strategies but still some potential limitations remain to be dissolved. Here, we worked out a simple cloning stra...
